I used to get hot spots when I would run in cycling socks. I used cycling socks because I know you can get hot spots from wearing socks that don't fit tight enough. But, I think I would get them because they were so thin, so it was basically the actually shoe that was rubbing and creating the hot spot. So maybe try a thicker sock, but still make sure it fits your foot tightly.
If you're already doing this, then I'm out of ideas. You might need some different shoes.
Shoes too tight? Is it from the friction or from circulation? If circulation, I'd try to loosen up the shoes. Too many people crank on those laces and pull pull pull. Not how it works. Try getting some Yankz! at your LRS for <$10. You won't go back to regular laces. I vote 'boo' on thick socks. Thick socks are sweat sponges and get all bunchy (highly technical, I know) and moisture and friction cause blisters. Go with microthin socks or no socks.

Try some body glide on the spot pre run...foot powder might help too. Something to cut the friction.
+1 on the thicker sock.
